Electrophoretic Characteristics of Macro Creatine Kinase Type 2 (Report of 3 Cases)




Abstract
Macro creatine kinase (CK) type 2(mitochondrial CK, CK-Mt) is an unusual isoenzyme of creatine kinase that migrates cathodically to CK-MM on electrophoresis and is an oligomeric mitochondrial CK. Its presence in serum has been associated with
metastatic
carcinoma and cirrhosis of the liver, the macro CK type 2 is composed of two forms of CK. One band has an electrophoretic mobility cathodal(oxidized form, fast fraction, oligomer, molecular weight : about 350, 000 daltons) to CK-MM and the other
has the
same mobility(reduced form, slow fraction, dimer, molecular weight : about 80, 000 daltons) as CK-MM. Macro CK type 2 is characteristical ly thermostable, not inhibited by antibodies to CK-M or -B ssubunits, and is dissociated by urea to show
same
mobility with CK-MM on electrophoresis.
We have confirmed these chracteristics of macro CK type 2, in the sera of 3 patients with lung cancer.
